Measuring the number and the area of pores of DPF filter

Greetings Dear friends:
I need to measure the number of pores (black dots) and the area of the pores from a SEM image of a diesel particle filter (DPF). I need your suggestions and codes. I need to know how to define for the program to choose only black dots with a certain size value.

Start off by trying to threshold for black blobs. See the Image Segmentation Tutorial and the interactive thresholding application in my File Exchange. You might need to do some background leveling first though.
But anyway, that will give you a start. You might need to mask off ROIs in the image. By the way, it doesn't much look like a TEM image, more like an SEM image to me.

Thanx for your kind reply.sorry i made mistake it was SEM, The TEM was for Soot particles .
